While each play only lasts 40 seconds, the clock stops at the end of incomplete passing plays and when a team calls a timeout. It also stops when a player goes out of bounds or if a penalty is called by an official.
If a player drops the ball or gets tackled while holding it, it’s a turnover. The team that recovers the ball gains possession of it and can continue to advance down the field. It’s possible for a team to turn the ball over without being tackled, however, as this is called a fumble.
